Well, you shouldn't have to tweak. Even I who is a techy guy dislikes tweaking wine. The only thing I've changed is I've enabled emulate virtual desktop in winecfg, which means I always run fullscreen apps in a window.
I just want you to install it, install the application you want to use. Then at the first sign of trouble, make a bugreport. That's all, just see if it works out of the box, if it doesn't you'll just have to use Windows.
Installing wine on a Mac might be a little trouble though since they don't seem to keep an official Mac download. Though there seems to be a guy that keeps on making Mac builds so there is an unofficial download.
